چکیده

The ubiquitin-proteasome system regulates protein degradation in every eukaryotic cell. Recent work has shown that protein turnover mediated by the ubiquitin-proteasome system plays a key role in synaptic plasticity.

Ubiquitination of Neurotransmitter Receptors and Postsynaptic Scaffolding Proteins

The human brain is made up of an extensive network of neurons that communicate by forming specialized connections called synapses. The amount, location, and dynamic turnover of synaptic proteins, including neurotransmitter receptors and synaptic scaffolding molecules, are under complex regulation and play a crucial role in synaptic connectivity and plasticity, as well as in higher brain functio...

Characterization of the Brain Proteasome and Its Interacting Proteins and Their Regulation by Neuronal Activity

Despite the importance of proteasome-mediated proteolysis in synaptic plasticity, protein quality control, and cell regulation, little is known about proteasome composition and regulation in the brain. This thesis represents the first detailed study of mammalian brain proteasomes. Using a new affinity purification method, 26S proteasomes were isolated from the cytosolic and the synaptic compart...

Ubiquitin and Protein Degradation in Synapse Function

The remodeling of synapses is a fundamental mechanism for information storage and processing in the brain. During brain development, and in response to learningrelated activity, synapses undergo remarkable structural changes, including growth, shrinkage, and elimination. Such structural plasticity provides a physical basis for enduring changes in neural circuits that are mediated by alterations...

Local ubiquitin-proteasome-mediated proteolysis and long-term synaptic plasticity

The ubiquitin-proteasome pathway (UPP) of protein degradation has many roles in synaptic plasticity that underlies memory. Work on both invertebrate and vertebrate model systems has shown that the UPP regulates numerous substrates critical for synaptic plasticity. Initial research took a global view of ubiquitin-protein degradation in neurons. Subsequently, the idea of local protein degradation...

Proteasome inhibition enhances the induction and impairs the maintenance of late-phase long-term potentiation.

Protein degradation by the ubiquitin-proteasome pathway plays important roles in synaptic plasticity, but the molecular mechanisms by which proteolysis regulates synaptic strength are not well understood. We investigated the role of the proteasome in hippocampal late-phase long-term potentiation (L-LTP), a model for enduring synaptic plasticity. We show here that inhibition of the proteasome en...
